The Three Mesquiteers - a group of cowboys consisting of Stony, Tucson, and Lullaby - team up to help George Brooke, the brother of Stony, who has been framed for murder. They soon discover a range war between cattlemen and sheepmen, with John Harvey, the head of Ashton Enterprises, attempting to put the cattle out of business. The Mesquiteers prove that the Brooke family estate is in Stony's name, stopping the sale and clearing George's name. Meanwhile, Tucson runs for sheriff and wins the election, exposing Harvey as the actual killer of Sylvia's father, allowing Stony to pursue a romance with Sylvia.
